Gluten, a protein found in wheat, barley, and rye, must be avoided by anyone with celiac disease or a gluten sensitivity. In many countries, foods that are labeled gluten-free must contain less than 20ppm of gluten. Additionally, wheat is a commonly-regulated food allergen around the globe.

When quantitative information is needed — in other words, when it is important to know precisely how much gluten is present — ELISA testing is the gold standard. This is true for gluten as well as other common food allergens.
ELISA, or Enzyme-Linked ImmunoSorbent Assay, is a type of test that requires specialized lab equipment and solid lab skills. Testing can take anywhere from 1-4 hours, but will return a precise value, usually measured in PPM, for the analyte in question. Because tests should be run in duplicate and alongside premade standards, ELISA testing only truly becomes cost-efficient when testing several samples at a time.

The high specificity of the G12 and A1 antibodies used in this test allows this method to precisely quantify gluten in food, beverage, and rinse water samples. Also suitable for surface monitoring.

GlutenTox® ELISA Rapid G12 holds AOAC certification for detecting gluten in foods, license number 042301. Validation studies showed no cross-reactivity (false positives) and no false negatives, as well as 100% specificity and 100% sensitivity for food testing. The test was validated against these Food Matrices: Soy flour, corn bread, seasoning mix, rolled oats, evaporated milk and incurred bread matrix.

The G12 antibody found in all GlutenTox test kits recognizes the 33-mer peptide. This is the toxic fraction of gluten in wheat, barley, rye, and even sometimes oat (Structural basis for gluten intolerance in celiac sprue, Science, 2002).

Of special note
Samples that have been subjected to excessive hydrolysis or fermentation may not be suitable for testing using this kit. This kit does not detect oat protein; however the ELISA Systems Oat Detection kit is suitable for that application.
